The isolation and properties of the proteins associated with tetanus antitoxic activity in equine plasma.

Several years ago, van der Scheer, Wyckoff, and Clarke (1) reported that antitoxic sera contained an electrophoretic component not present in normal horse sera. This component which was designated as T-globulin had a mobility between the pand r-globulins. Kekwick and Record (2) found that in diphtheria antitoxic sera the immune activity was present in both 0 and y components. These globulins were separated electrophoretitally, and found to be different in their flocculation times and in their ratios of in vivo to in vitro activities. From the results of these and other investigators, it is evident that certain antigens injected into the horse produce antibodies associated with more than one serum component. It has already been reported from this laboratory (3) that the cow also produces antibodies associated with y and T components. Both of these components were isolated from hyperimmune plasma in electrophoretically homogeneous state by means of the alcohol fractionation procedures devised by Cohn and his collaborators (4,5). By these same methods, some studies have been made on tetanus antitoxic plasma from the horse. It’ was found that homogeneousr-globulin and nearly homogeneous T-globulin could be isolated. Both of these proteins have been characterized by physical and chemical studies. In addition to the yand T-globulins, however, there is present in the plasma of the horse a large quantity of additional material with which antitoxic activity is associated. Although this protein has not yet been isolated in electrophoretically homogeneous form, it appears to be associated with the water-soluble (pseudoglobulin) P-globulin fraction.
